Below are recipes for culturing Photobacterium species. They may be for a specific species, or for the genus in general. Remember, all Photobacterium require salt to grow. This is because they are a primarily marine genus and have adapted to a salty environment. The Media:

Glycerol Chalk Media
NOTES: Add enough distilled water to bring the total volume to 1 L. Mix these components well by stirring and heating gently. This media should be autoclaved for 15 min. at 15 psi and 121 C. After autoclaving, pour into dishes as soon as its cool enough to handle. The CaCO3 may come out of suspension while pouring - keep stirring! This media was designed for any Photobacterium sp. and also Lucibacterium sp. |
Luminous Medium
NOTES: Add enough distilled water to bring the total volume to 1 L. Mix these components well by stirring and heating gently to a boil. This media should be autoclaved for 15 min. at 15 psi and 121 C. After autoclaving, pour into dishes as soon as its cool enough to handle. The CaCO3 may come out of suspension while pouring - keep stirring! This media was designed for any Photobacterium sp. and also Alteromonas hanedai, Shewanella hanedai, Vibrio sp. |
Photobacterium Agar
NOTES: Add enough distilled water to bring the total volume to 1 L. Mix these components well by stirring and heating gently. This media should be autoclaved for 15 min. at 15 psi and 121 C. After autoclaving, pour into dishes as soon as its cool enough to handle. The CaCO3 may come out of suspension while pouring - keep stirring!Final pH should be 7.2 +/- 0.2 at room temperature (25 C). This medium can also be a broth (Photobacterium broth - different from below). To do this, don't add the agar and prepare the same way as mentioned. Surprisingly, despite the name of the medium, this media was designed for the bacterium Acetobacter pasteurianus. |
Photobacterium Broth
NOTES: Add enough distilled water to bring the total volume to 1 L. Mix these components well by stirring and heating gently. This media should be autoclaved for 15 min. at 15 psi and 121 C. Final pH should be 7.0 +/- 0.2 at room temperature (25 C). This medium can also be bought from the Company Difco Laboratories as a pre-mixed powder of dry ingredients. This broth is used to show luminescence of the cultured organisms and also for maitenance of Photobacterium phosphoreum and also Alteromonas hanedai, Shewanella hanedai, and Vibrio sp. |
Photobacterium MPY Medium
NOTES: Add enough distilled water to bring the total volume to 1 L. Mix these components well by stirring and heating gently. This media should be autoclaved for 15 min. at 15 psi and 121 C. Final pH should be 7.4 +/- 0.2 at room temperature. This broth is used to grow and maintain Photobacterium leiognathi. |
Seawater Agar
NOTES: Mix beef extract, peptone, and distilled water to a volume of 250 ml. Mix well. Make sure pH is 7.8. Then, boil for 10 minutes. Bring the total to 1 L by adding the Seawater (natural or artificial) and mixing. Readjust the pH to 7.3. Add the agar and boil further. Pour hot solution into culture tubes or other container for autoclaving. Autoclave at 15 psi pressure and 121 C for 15 minutes. Redistribute if necessary into petri plates or other vessels. This agar media is used to grow and maintain Aiteromonas rubra, Brevibacterium stationis, Chromonalobacter marismortui, Flectobacillus marinus, Marinococcus albus, Marinococcus halophilus, Pastuerella piscicida, Photobacterium phosphoreum, Planococcus citreus, Planococcus kocurii, and assorted Vibrio species. |
